pCSG-IBAwt2 is a large expression vector with universal features for transient expression as well as for generation of stable mammalian cell lines. The vector carries an ampicillin resistance cassette for selection of transformed E. coli cells and ColE1 origin for a high plasmid copy number. Extrachromosomal replication in mammalian cells could occur either by origin of replication from Epstein-Barr Virus (oriP) or by SV40 ori. For the former the vector provides the EBNA-1 gene and for the latter the cell line has to be latently infected with SV40 or express the SV40 large T antigen (e.g. HEK293T, COS-1, COS-7). Stable cell lines can be selected by the neomycin resistance gene (NeoR). In addition, the human cytomegalovirus (CMV) immediate-early promoter enables a high-level expression. During translocation from the cytosol the signal peptide is removed from the protein by endogenous proteases.

The onset of angiogenesis is believed to be an early event in tumorigenesis and may facilitate tumor progression and metastasis. Several growth factors with angiogenic activity have been described. These include FGF, PDGF, vascular endothelial growth factor (VEGF), VEGF-B, VEGF-C, VEGF-D (or FIGF), EG-VEGF and placenta growth factor (PlGF). VEGF-D undergoes a complex proteolytic maturation, generating multiple processed forms which bind and activate VEGFR-2 and VEGFR-3 receptors. This protein is structurally and functionally similar to vascular endothelial growth factor C.

Duplex DNA is isolated from bacteriophage lambda (cI857ind 1 Sam 7). The bacteriophage is grown in an E. coli host which is deficient in adenine methylase (dam-). Therefore, those deoxyadenosine positions which are methylated in a wild type E. coli host are not methylated in the dam- host. This DNA gives complete digests with restriction endonucleases that are sensitive to dam methylation. Lambda DNA (dam-) is 48,502 base pairs in length.

The ATF/CREB family consists of a series of transcription factors that function by binding to the cAMP responsive element (CRE) palindromic octanucleotide, TGACCTCA. The best characterized members of this gene family include CREB-1, CREB-2, LZIP (also designated CREB3 and Luman), CREM-2, ATF-1, ATF-2, ATF-3, ATF-4, ATF-6 and ATF-7.
